It's actually decribed on Page 23 of the manual.

The "Camera ROI..." menu item allows you to specify a Region Of Interest on the Camera's CCD to image with. This item will only be selectable if the Camera Video Format: menu is set to a Format 7 (i.e. FMT7) video format, the Format 7 mode you selected allows ROI and if the camera is not currently displaying video.

So my guess is that you selected Format 7, then clicked "Start Video" and then can not access the ROI menu item. What you need to do is select Format 7, select the ROI item, fill in the ROI area and then click "Start Video".
