Say I`m photographing NGC 2516 for instance which according to Equinox 6 has a Declination of; -60 degrees, 52 minutes & 1.2 seconds won`t the Easy guider`s second camera be positioned diametrically opposite this (180 degrees distant)?

Nope, the Guider orientation doesn't make any difference. Astro IIDC wants the absolute declination of the object, so enter -60 or -61 degrees. You don't have to be precise and a few degrees won't make a large difference either.

Secondly the idea behind entering the Dec is that you can do the calibration on some easily found brighter star, then you can move to the actual target, enter it's declination (so Astro IIDC can compensate for the changed in declination) and just start guiding on it. This works best with a secondary scope for guiding, because you won't be rotating the camera or changing orientation like you would with an Off Axis Guider when your hunting for guide stars.
