I finally got back yesterday and now have enough bandwidth here to look at your image.
I don't know where that large mark is.
If you have a high power magnifier, you may be able to tell if it's a divot / pit on the outer glass cover or a clump of dust on the CCD glass cover itself. Basically just shift the focus of the lens from one surface to the other.
